What is Hirudin peptide powder?
Hirudin peptide is a peptide thrombin inhibitor designed using hirudin as a template. The number and sequence of its amino acids also vary depending on the designer. The original hirudin peptide was designed by Margzncre et al. It has 20 amino acid residues It contains two structural domains and a linking region. The C-terminal is derived from the carboxyl terminus of hirudin and can tightly bind to the catalytic reaction site of thrombin. The design of leech peptide consists of three parts: hirudin carboxy-terminal 10 peptide, the specific recognition sequence of the thrombin catalytic active site and the functional connection bridge between the two. Leech peptide directly binds to the thrombin active site through the (d-Phe)-Pro-Arg-Pro sequence. If 1-Phe is used to replace the N-terminal d-Phe, the anti-thrombin activity of the leech peptide will be greatly reduced; when When all the (d-Phe)-Pro-Arg-Pro sequences are removed, no inhibition of thrombin will be detected when the concentration is no higher than 10 μM. The mechanism of action of hirudin on thrombin is not completely the same as that of hirudin, and its half-life will also be different. In addition, in vitro experiments also show that hirudin has a stronger inhibitory effect on thrombin bound to fibrin (blood clot) than hirudin.
