What is ferulic acid Powder?
Ferulic acid, whose chemical name is 3-methoxy-4-hydroxycinnamic acid, is one of the derivatives of cinnamic acid. Its content is found in ferulic acid, angelica root, Ligusticum chuanxiong, Cimicifuga, jujube kernel and other Chinese medicinal materials. Ferulic acid has two isomers, cis and trans, both of which are light yellow solids. Ferulic acid extracted from plants can be obtained from plants in three ways: one is obtained from the combination of ferulic acid and some small molecules, the other is obtained from plant cell walls, and the third is obtained through tissue culture. . The synthetic ferulic acid is usually obtained through chemical reaction using vanillin as the basic raw material.

What is the role of ferulic acid?
Ferulic acid, also known as Ferulate, has multiple functions.
1. Anti-radiation effect
Radiation-induced organ failure is largely caused by chronic peroxidative damage. Radiation damage to the body is divided into two types: direct damage and indirect damage. Direct damage means that radiation directly causes the breakage of some sensitive molecules in cells; indirect damage causes radiolysis of water, which leads to an increase in intracellular reactive oxygen species and then causes subcellular structures. changes, therefore antioxidants are widely used in the treatment of radiation damage. Protection of cells from reactive oxygen species requires the maintenance of endogenous sulfhydryl pools in cellular homeostasis, particularly the stabilization of glutathione (GSH) and nicotinamide adenine dinucleotide phosphate (NADPH) levels. Glutathione provides reducing equivalents for the conversion of hydrogen peroxide and lipid peroxides into water and lipid alcohols, protecting sulfhydryl protein groups from oxidative damage. The rate-limiting reaction of glutathione biosynthesis is catalyzed by glutamate-cysteine ligase (GCL), which consists of a catalytic subunit (GCLC) and a regulatory subunit (GCLM) . Nicotinamide adenine dinucleotide phosphate is an important antioxidant component in tissues and can maintain the redox potential of cells by reducing the reducing equivalents of glutathione reductase and thioredoxin.
As a phenolic plant component, ferulic acid has strong antioxidant activity and greatly promotes human health. Ferulic acid can significantly increase the levels of glutathione and nicotinamide adenine dinucleotide phosphate in irradiated cells, and has a protective effect on irradiated endothelial cells. Heme oxygenase is an enzyme with antioxidant effects. It can convert heme into biliverdin and finally into the antioxidant bilirubin. Ferulic acid can well regulate the expression of this enzyme. , thus playing a protective role in radiation protection.
2. Antioxidant function
Metabolism is a characteristic of living organisms. At the same time, living organisms are constantly being attacked by reactive oxygen species (molecules or free radicals that are directly or indirectly converted from molecular oxygen and are more active than molecular oxygen), free radicals (also known as free radicals, Refers to the attack of atoms, atomic groups or molecules in a special state containing unpaired electrons in the outer orbit. Both of these substances can directly participate in the formation of tumors or induce the production of carcinogens, causing changes in the DNA of living organisms and proto-oncogenes. Start, thereby promoting the body to produce cancer cells and reducing antioxidant damage, which is a key link in preventing and treating related diseases. Studies have shown that ferulic acid can destroy free radicals and restore normal functions of living organisms. Ferulic acid can inhibit the enzymes that generate free radicals in living organisms. On this basis, it can also increase the enzymes that scavenge free radicals. At the same time, ferulic acid can greatly enhance the activity of aldehyde reductase and glutamic acid transsulfurase, and control the proportion of active tyrosinase. Research shows that ferulic acid has significant antioxidant effects and has a good scavenging effect on ammonia peroxide, superoxide radicals, hydroxyl radicals, nitro peroxide, etc.
3. Antibacterial and antiviral functions
The study found that after the macrophages of experimental mice were infected with influenza virus, a blank control was set without treatment, and the test group was treated with ferulic acid and isoferulic acid. Analysis of the results showed that the production of interferon decreased faster in the test group. In recent years, there have been many reports on the significant inhibitory effects of ferulic acid on cold viruses (IV), respiratory syncytial virus (RSV) and HIV (HIV). The effects of ferulic acid on inflammatory proteins were studied in the same cell line. As a result, ferulic acid can drastically reduce the production of this protein. Among them, ferulic acid has an inhibitory effect on HIV, so there is a possibility that ferulic acid will become a chemotherapeutic agent in the future. It is speculated that the inhibitory mechanism of ferulic acid on viruses is related to its ability to reduce the activity of xanthine oxidase, because this type of enzyme can generally cause some inflammation. It is speculated that the antibacterial function of ferulic acid is mainly due to its effect on bacteria. N-acetyl transferase has a strong inhibitory effect.

There are also two main processes for the preparation of ferulic acid. The first one is mainly based on plant extraction. Ferulic acid in plants is mostly cross-linked with polysaccharides and lignin through ester bonds or is itself esterified or etherified to form diferulic acid. Generally, Use alkaline and enzymatic methods to break the ester bond to release ferulic acid, and then use a suitable solvent for extraction.
1. Alkaline solution
Ferulic acid from the cell wall can be released by reacting with 4% sodium hydroxide at room temperature for 24 hours under nitrogen atmosphere. Recent research has found that by increasing the extraction temperature and adding suitable protective agents, most of the ferulic acid in wheat bran can be freed in a short period of time. Using a low-concentration sodium hydroxide solution can release most of the ferulic acid in wheat bran at an appropriate extraction temperature. Adding sodium sulfite during the extraction process can increase the recovery rate of ferulic acid. Due to the complex composition of the alkali solution, especially the pigment substances, currently, the separation method of ferulic acid in the alkali solution mainly uses the activated carbon adsorption method. Oryzanol contains the structural unit of ferulic acid, which exists in the form of ester and is easy to decompose. Therefore, you can first hydrolyze oryzanol with alkali and then use acidification to prepare ferulic acid. The reaction formula of oryzanol is to hydrolyze oryzanol to prepare ferulic acid. Convenient, the yield is as high as 85.7%, and the by-product is cyclopineol. Moreover, oryzanol has wide sources, large output, and moderate price.
2. Ferulic acid esterase method
Ferulic acid esterase is an enzyme that can liberate ferulic acid from methyl ferulate, oligosaccharide ferulic acid ester and polysaccharide ferulic acid. Fungi, bacteria, and yeast all secrete feruloyl esterase. Using Aspergillus niger as the strain, a liquid submerged fermentation method was used to prepare a mixed enzyme preparation containing ferulic acid esterase and arabinoxylanase. The mixed enzyme preparation was used to act on the destarched wheat bran. It was found that after three times of degradation, The wheat bran degradation rate reaches 55.46%.
3. Plant tissue culture method
Plant tissue culture is an important way to obtain ferulic acid. Some studies have shown that culturing certain plant tissues can produce higher yields of ferulic acid derivatives. For example, cell suspension culture of sugar beet and corn can obtain water-soluble ferulic acid glucose ester, ferulic acid sucrose ester, etc., and the content can be as high as 20.0 μmol/g callus (dry weight). In the direct extract, the ferulic acid content is relatively low and requires further purification.

The chemical synthesis method usually uses vanillin as the basic raw material, and the main organic reactions used are Wittig-Horner reaction and Kneoevenagel reaction.
1. Wittig-Horner reaction to synthesize ferulic acid
The Wittig-Horner reaction occurs between triethyl phosphite acetate and acetyl vanillin in a strong alkaline system, and then acidified with concentrated hydrochloric acid to obtain ferulic acid. This method requires pre-protection of the phenolic hydroxyl group, otherwise due to the presence of a strong base, the formation of sodium phenolate will inhibit the reaction between the carbonyl group and the carbanion, and side reactions may easily occur to generate impurities.
2. Kneoevenagel reaction to synthesize ferulic acid
A small amount of organic base is added to the pyridine solvent as a catalyst. Kneoevenagel reaction occurs between vanillin and malonic acid to generate ferulic acid. The catalysts include piperidine and aniline. However, the reaction time of this method is long, up to three weeks, and a mixture of trans and cis ferulic acid is obtained.
3. Biosynthesis method
The biosynthesis method uses several microorganisms to convert ferulic acid precursors into ferulic acid, such as converting eugenol cinnamic acid ester extracted from clove oil into ferulic acid. Biosynthesis is a clean and effective synthesis method, but a mass production method has not yet been developed.
